Total twenty five organic solvent tolerant bacteria (OSTB) have been isolated from six different sites. Polyphasic approach was employed for identification of OSTB including morphological and biochemical characterization, fatty acid profiling and 16S rRNA gene sequencing. Amongst them, twenty belonged to Bacillus spp., two to Micrococcus sp., and one each of Geobacillus sp., Terribacillus sp., and Staphylococcus sp. Thus, Bacillus group of species are dominating in this study. With this approach we could identify 19 isolates up to species level while six OSTB could be identified only up to genus level (Bacillus). Hence, future study in this direction using the latest techniques like DNA-DNA hybridization would lead to proper identification of these isolates and might help in establishing the relationship with known Bacillus spp.en_US
